Abstract
Background:
@EN There have been a lot of trials for understanding the role of biliary protein in gallstone formation mainly cholesterol stone. However, the exact roles has not been completely documented in pigment gallstone which shows high incidence in the
Orient.
@ES Methods:
@EN We tried to extract, isolated and characterized proteins in patients with gallstones. 10 pigment gallstones were obtained at cholecystectomy and extracted with ethanol/ether. Then, isoelectric focusing were performed and gallstone proteins
were
analyzed by SDS-polyacrylamide gel electrophoresis. The amino acids were also analysis by autoamino acid analyzer system.
@ES Results:
@EN 1) The mean amount of gallstone protein was 16.18mg%.
2) The protein showed major band at low pH on isoelectric focusing.
3) On SDS-PAGE, low molecular proteins bands were noticed; 90% showed below 45Kda.
4) The composition of aspartic acid, glutamic acid: 22.7%
@ES Conclusion:
@EN We could suggest that the proteins in black pigment gallstones are mainly low molecular weight with acidic proteins and these proteins could play an important role in the formation of gallstones.
